The suitability of a fluorescence microscope depends on the fluorophore,
excitation/emission requirements, specimen preparation and imaging workflow.
Customers should provide the intended fluorescence application before ordering
so that the optical and filter configuration can be reviewed.
Trinocular Observation System
The microscope is equipped with a 30° inclined trinocular observation head.
The trinocular configuration supports visual observation while also providing a
dedicated optical path for compatible imaging equipment when required.
Standard wide-field eyepieces are WF10X with a
22 mm field number, providing a wide observation field for routine
microscopic examination.
Infinity-Corrected Objective Configuration
The UMMS013 objective set includes plan achromatic and fluorescence-oriented
objectives for routine transmitted-light and fluorescence applications.
| Objective | Numerical Aperture | Working Distance | Configuration |
|---|---|---|---|
| PL 4X | 0.10 | 19.8 mm | Plan achromatic |
| PL 10X | 0.25 | 5.0 mm | Plan achromatic |
| PL FL 40X | 0.85 | 0.42 mm | Spring-loaded fluorescence objective |
| PL 100X | 1.25 | 0.36 mm | Spring-loaded oil-immersion objective |
With WF10X eyepieces, the corresponding nominal total magnifications are
40X, 100X, 400X and 1000X.
Reflected Fluorescence Illumination System
The reflected fluorescence system uses a 100W DC mercury lamp
with a separate power supply configuration. Power-box voltage should be confirmed
according to the destination market and supplied configuration.
A rotary fluorescence reflector provides four excitation channels:
UV, violet, blue and green.
| Channel | Excitation Wavelength | Emission Wavelength |
|---|---|---|
| UV | 320–380 nm | 435 nm |
| Violet | 380–415 nm | 475 nm |
| Blue | 450–490 nm | 515 nm |
| Green | 495–555 nm | 595 nm |
Before ordering, the required fluorescence dyes or fluorophores should be provided
so that the available excitation and emission filter combination can be checked
against the intended application.
Mechanical Stage and Focusing System
UMMS013 uses a 210 × 140 mm double-layer mechanical stage
with an X-Y movement range of 75 × 50 mm.
The mechanical control system allows precise positioning of prepared microscope slides.
Coaxial coarse and fine focusing is provided, with a fine-focus graduation of
2 μm. Locking and limit mechanisms are incorporated into the
focusing system to support controlled operation.
Transmitted Brightfield Illumination
In addition to reflected fluorescence observation, UMMS013 includes a transmitted-light
system for conventional brightfield microscopy.
The transmitted illumination configuration includes:
- Abbe condenser NA 1.25
- Adjustable condenser height
- Blue filter
- Frosted glass
- 6V 30W halogen lamp
- Adjustable illumination brightness
- Built-in field diaphragm
